One of the most important properties to consider when engineering biologics is protein solubility. Poor solubility affects the yield as a result of low expression in production, or problems in purification, degradation, storage, and/or administration.
This application brief, "Modeling Protein Solubility", written by Velin Z. Spassov, describes two recently published in silico methods for evaluating protein solubility and their utility in engineering of biologics. Unlike most of the existing methods, these methods account for the energy of protonation of acidic and basic amino acid residues.
